草甘膦抗性杂草常通过靶基因EPSPS扩增进化,但非EPSPS相关染色体结构异常未见报道。本研究发现抗草甘膦Amaranthus tuberculatus中存在约 6 μm 的大染色体(正常 2—3 μm),由着丝粒周围异染色质扩展和 5S rDNA 重复形成,无EPSPS且不传递给后代,为抗性染色体进化研究提供新方向。
结果发现,转基因烟草同时表达gr79 epsps和gat基因对草甘膦的抗性显著高于gr79 epsps或gat单基因烟草。 进一步利用农杆菌介导法将GR79 EPSPS-GAT双基因载体转化棉花,利用多种分子检测手段,获得含有一个和两个EPSPS-GAT表达盒插入的转基因棉花GGCO2和GGCO5,且GR79 EPSPS和GAT基因在转录和翻译水平均高表达。
